Miriam Shor moved to New York City and established her stage career, notably starring in Hedwig and the Angry Inch as Yitzhak. She reprised the role in the film adaptation Hedwig and the Angry Inch (2001). Her early screen credits include the sitcom Then Came You (2000), the comedy film Bedazzled (2000), and the television series Inside Schwartz (2001–2002). She also guest-starred on My Name Is Earl, The West Wing, and later appeared in Shortbus (2006).
In 2006, Shor starred in the comedy series Big Day. She appeared in The Cake Eaters (2007) and starred in the drama series Swingtown (2008). Her television work also includes recurring roles on Damages and appearances on Law & Order: Criminal Intent, Bored to Death, and Royal Pains. In 2011, she portrayed Anna in the HBO miniseries Mildred Pierce.
Shor starred as Cricket Caruth-Reilly in the comedy-drama series GCB (2012) and had a recurring role as reporter Mandy Post on The Good Wife beginning in 2012. She also appeared in Broadway Bares in 2012. In 2014, she was cast as a series regular on Younger, which premiered in 2015 and ran for seven seasons. For her performance, she received a Critics' Choice Television Award nomination for Best Supporting Actress in a Comedy Series in 2018. That same year, she made her directing debut with an episode of the series.
Her stage work includes portraying Madelaine True in The Wild Party at New York City Center in 2015. She had a recurring role as Erica Haskard in the final season of The Americans, earning a Screen Actors Guild Award nomination as part of the ensemble cast. Shor later appeared in the film Lost Girls (2020), the comedy-drama Magic Hour, and the science fiction film The Midnight Sky (2020), directed by George Clooney.
In 2023, Shor portrayed Cynthia O'Neal in Bradley Cooper's Maestro and appeared as Recorder Vim in Guardians of the Galaxy Vol. 3. That same year, she appeared in American Fiction and guest-starred on And Just Like That.... In 2025, she had a recurring role in the Apple TV+ science fiction series Pluribus.
In 2026, Shor directed and produced the documentary My NDA alongside Juliane Dressner. The film premiered at the 2026 South by Southwest Film & TV Festival.
