Jane Seymour began her acting career in 1969 with an uncredited appearance in Richard Attenborough's film Oh! What a Lovely War. She took on her first major film role in the war drama The Only Way (1970), playing Lillian Stein. In 1973, she gained a major television role as Emma Callon in the series The Onedin Line. That same year, she appeared as Prima in the television miniseries Frankenstein: The True Story and portrayed Pamela Plowden, Winston Churchill's girlfriend, in Young Winston.


Seymour gained international recognition in 1973 for her portrayal of Solitaire in the James Bond film Live and Let Die. She later played Princess Farah in Sinbad and the Eye of the Tiger (1977), the third installment of Ray Harryhausen's Sinbad trilogy. In 1978, she appeared as Serina in the Battlestar Galactica film and the first five episodes of the television series. She subsequently starred opposite Chevy Chase in the comedy Oh Heavenly Dog.

In 1980, Seymour made her Broadway debut as Constanze in Peter Shaffer's play Amadeus, appearing alongside Ian McKellen and Tim Curry. That same year, she played Elise McKenna in the period romance Somewhere in Time, opposite Christopher Reeve. She appeared in the television film East of Eden (1981), portraying Cathy Ames and earning a Golden Globe Award for her performance. She subsequently starred in The Scarlet Pimpernel (1982) with Anthony Andrews and Ian McKellen, and appeared in Lassiter (1984), opposite Tom Selleck.

In 1988, Seymour starred as Natalie Henry in the television miniseries War and Remembrance, the continuation of The Winds of War. She also portrayed opera singer Maria Callas in the television film Onassis: The Richest Man in the World (1988), earning an Emmy Award. In 1989, she appeared in the television film La révolution française, portraying Marie Antoinette.

During the 1990s, Seymour gained recognition for her leading role as Dr. Michaela "Mike" Quinn in the television series Dr. Quinn, Medicine Woman. She reprised the role in the television films Dr. Quinn, Medicine Woman: The Movie (1999) and Dr. Quinn, Medicine Woman: The Heart Within (2001). Her work on the original series earned her a second Golden Globe Award.

Seymour continued working primarily in television during the 2000s. In 2004 and 2005, she made six guest appearances on the WB series Smallville as Genevieve Teague. She appeared in the comedy Wedding Crashers (2005), playing Kathleen Cleary opposite Christopher Walken. In 2006, she appeared in the short-lived series Modern Men, guest-starred on How I Met Your Mother and Justice, and in 2007, she guest-starred on In Case of Emergency. She also appeared in ITV's Marple: Ordeal by Innocence and competed on the fifth season of Dancing with the Stars in 2007, finishing sixth with professional partner Tony Dovolani. She subsequently guest-starred in the ABC crime dramedy Castle.

Seymour appeared in the Hallmark Channel film Dear Prudence (2008), the romantic comedy Love, Wedding, Marriage (2011), and the Hallmark Movie Channel film Lake Effects (2012). In April 2016, she starred as Florence Lancaster in Noël Coward's play The Vortex, presented in Singapore by the British Theatre Playhouse.

In 2020, Seymour starred in the Australian comedy-drama Ruby's Choice, directed and produced by Michael Budd. She portrayed the title character, a woman living with early dementia. Her performance earned her the Australian screen industry Network Award for Best Actress. The film was released theatrically in Australia and New Zealand on March 3, 2022, and premiered in the United States at the Santa Barbara International Film Festival on March 7, 2022. It later received a U.S. premiere at the Burbank International Film Festival, where it won Best Foreign Film and Best Feature Film. Ruby's Choice was released nationwide in North America on May 7, 2024.

In 2022, Seymour began playing the title role in the Irish Acorn TV series Harry Wild. She appeared in the Netflix film Irish Wish, released on March 14, 2024. In 2025, she began hosting the Acorn TV and BBC America series Relative Secrets.
